REAGEN   The Domoic Acid ELISA is a competitive enzyme immunoassay for the quantitative and sensitive detection of Domoic Acid. Domoic Acid is one of the toxins associated with amnesiac shellfish poisoning (ASP). This test is suitable for the quantitative and/or qualitative detection of Domoic Acid in water samples as well as shellfish samples.

Procedure Overview

REAGEN  The method is based on a competitive colorimetric ELISA assay. The ASP antigen has been coated in the plate wells. During the analysis, sample and ASP antibody (Antibody#1) and HRP-Conjugate (HRP-Conjugated Antibody #2) are added to the wells for incubation. If the ASP residue is present in the sample, it will compete with the antigen on the plate wells for the antibody. The secondary antibody, tagged with a peroxidase enzyme, targets the primary antibody that is complexed to the drug coated on the plate wells.The resulting color intensity, after addition of substrate((TMB), has an inverse relationship with the target concentration in the sample. The color reaction is stopped after a specified time and the color is evaluated using an ELISA reader. The concentrations of the samples are determined by interpolation using the standard curve constructed with each run.

Kit Contents, Storage and Shelf Life

REAGEN  Domoic Acid ELISA Test Kit has the capacity for 96 determinations or testing of 42 samples in duplicate (assuming 12 wells for standards). Return any unused microwells to the foil bag and reseal them with the desiccant provided in the original package. Store the kit at 2-8°C *. The shelf life is 12 months when the kit is properly stored.

*  If you are not planning to use the kit for over 1 months, store Antibody Solution and HRP-Conjugated at -20°C or in a freezer.
